Franklin County comprises the Chambersburg–Waynesboro, PA metropolitan statistical area, which is also included in the Washington–Baltimore combined statistical area. The county is part of the South Central Pennsylvania region of the state.

Guilford is located in eastern Franklin County at 39°55′10″N 77°35′54″W (39.919506, -77.598262). It is primarily in Guilford Township, but a portion extends north into Greene Township. The area is also known as Guilford Hills.

U.S. Route 30 passes through the community, leading east 2 miles (3 km) to Fayetteville and west 4 miles (6 km) to Chambersburg, the county seat.

Guilford Township was originally known as Township Number 14, Range 14W of the Connecticut Western Reserve, and was one of the first townships organized after the creation of the county in 1818.
