In 1818, a dispute about the recording of deeds prompted residents of the two villages to secede from Pittsford and form the town of Henrietta.

Of this total, 35.4 square miles (92 km2) is land and 0.2 square miles (0.52 km2) (0.53%) is water. Both the New York Thruway (Interstate 90) and Interstate 390 run through Henrietta. A short section of the Erie Canal passes through the northeastern side of the town.
